While the findings hint at a possible role for oral bacteria in brain inflammation, the study stops short of proving causation, emphasizing that Alzheimer’s remains a complex condition involving multiple biological factors.
Gum Bacteria and Brain Inflammation: A Possible Mechanism
The study proposed that P. gingivalis could enter the brain via the bloodstream, where its toxic enzymes—gingipains—may damage proteins and trigger inflammation, accelerating neuronal damage. Experiments in mice showed that oral infection with the bacterium led to increased amyloid beta production, a hallmark of Alzheimer’s, and neuronal loss. Blocking gingipains reduced these effects, suggesting a potential therapeutic target.

ImP’s role in promoting the buildup of amyloid and tau proteins—key features of Alzheimer’s—mirrors the effects of gingipains. The study’s authors suggested that targeting ImP could offer a new strategy for reducing dementia risk, though human trials are still in early stages.

The review, which included data up to March 2025, concluded that gum disease and tooth loss were significantly linked to Alzheimer’s risk. However, the authors cautioned that the studies were observational, and confounding factors—such as lifestyle and genetics—could influence the results.

Despite these findings, the scientific community remains cautious. Calling it the proven cause of Alzheimer’s would go far beyond the available evidence.
